How does this recalibrate? Do you have to buy something to recalibrate the scale
For the calibration of this pet scale:Press the power button: after ON/OFF displays PE-400, press ON/OFF+MODE at the same time to display E-100, then press MODE to display E-50, and then press MODE to display E-25. At this time, 25kg weight is put ON for calibration. Show Good calibration complete.

Does the weight change as weight is added or does it “lock” on number until you take weight off and let it go back to zero? Thank you
This product can switch the locking mode by pressing the C/R button. Press the button to manually lock. To manually lock, you need to press the HOLD button to lock the number displayed on the instrument. If it is in automatic locking mode, the instrument will automatically lock the weighing value after the heavy load or load is stable. When locked, the display number remains unchanged, and the unit (Kg or LB) flashes.

Can you buy a replacement screen?
I do not know if you can or not but this scale wasn't what it was promoted to be.

My scale turns on and when you put weight on it it will only read 0. I have changed the batteries . Any help?
My son had to reset ours a few times and then start with smaller weight. It will also read zero if the weight isn't evenly distributed between both ends.

Will it work for a great dane
Yes, it would work good. We used it for show hogs. I would say depending on where you keep it, we put the no skid tape on it to prevent slipping for our animals.
